D7.2 Software to define tumour subclones and association with therapy response
Flow cytometry is an important diagnostic tool in childhood acute lymphoblastic leukaemia (ALL), flow cytometry data analysis is limited by multiple sources of bias and variation. We present a unified machine learning framework for automated analysis of a standardized diagnostic paediatric leukaemia staining that can overcome these challenges. We applied our framework in a large cohort of ALL flow cytometry samples and demonstrated how it can robustly extract the frequencies of cell lineage populations with minimal expert intervention. This work provides a proof of concept that our method meets the needs of an automated analysis tool for diagnostic flow cytometry data.

Oncogene-driven metabolic rewiring in cancer is key to allow proliferation of tumour cells in low nutrient and oxygen conditions. To study such phenomena, reconstructing context-specific metabolic models through omics data integration is crucial. Here we report the original pipeline to construct context-specific metabolic models from scRNA-seq data and we applied it to scRNA-seq data from Ewing Sarcoma.

D4.3 Topological analysis of multi-omics and multi-cancer molecular networks resulting in the definition of molecular mechanisms
Three types of network-based analysis of gene-gene interaction networks have been suggested and tested on the multi-omics paediatric cancer datasets. User-friendly computational environment for joint application of matrix factorization and network analysis has been implemented.

D1.3 Synthetic data for testing and training patient, cancer, and drug models
Synthetic data generation is emerging as an important solution for precision medicine. Therefore, an explainable Variational AutoEncoder (VAE) model is developed for synthetic transcriptomics data generation in medulloblastoma. The model can be used to complement and interpolate available data with synthetic instances. It is also transparent as it is able to match the learned latent variables with unique gene expression patterns. The model can also be adapted to other pediatric cancers and the resulting synthetic datasets used to test and train patient, cancer, and drug models in other work packages of the iPC project.

With the development of the NaviCell 3.0 web server, there is a complete and automated web-based infrastructure for hosting molecular maps, patient similarity network maps, and multi-omics datasets for the project. The NaviCell platform supports molecular map navigation and exploration using the Google maps engine. The logic of navigation is taken from Google maps. This NaviCell 3.0 web-server is freely available and several step-by-step tutorials are accessible.

D7.1 Application of software enabling computational deconvolution of bulk RNA-sequencing data to immune cell profiles of patient samples
Computational deconvolution of bulk RNA-sequencing data to infer cell type composition of a sample is challenging. Benchmarking of various computational deconvolution tools revealed various data processing parameters that impact deconvolution accuracy and revealed the importance of a complete reference matrix. As a complete reference matrix is often not available, an algorithm was designed that can handle missing cell types. This algorithm can be applied to establish the immune cell repertoire of primary tumor biopsies without prior knowledge of the full spectrum of cell types in the biopsy.

In this deliverable, demographic, clinical, and molecular profiles were collected for several pediatric and adult tumors. In addition, the focus here is on collections of single cell profiles of high risk cancers. The datasets will be used to evaluate the effects of treatments and perturbations on cancer cells, build models, and provide information on deciphering regulatory interactions. These data will allow characterization of cancer cell types that predict treatment outcome, as well as cell types that are resistant to therapies.

The EU-funded 6GTandem project started in 2023 and is now coming to a close. Its mission: to explore a dual-frequency approach that combines the strengths of sub-THz connections with those of sub-10 GHz systems.
This demonstrator tackles a key bottleneck: moving massive amounts of data within dense networks. In the lab, researchers showcase an ultra-high-capacity link using Polymer Microwave Fiber and a wireless sub-THz connection - reaching 100 Gb/s over 1 m of PMF and 24 Gb/s wirelessly.

The EU-funded 6GTandem project started in 2023 and is now coming to a close. Its mission: explore a dual-frequency distributed MIMO architecture for future 6G networks, combining reliable sub-10 GHz links with ultra-fast sub-terahertz connections.
This demonstrators realistic scenario - an industry hall – addresses a key 6G challenge: delivering reliable, high-capacity coverage in complex indoor environments. At its core is a Central Unit (CU) that generates and processes signals, distributing them via plastic microwave fibers—Radiostripes—along the ceiling to multiple Radio Units.
These Radio Units can transmit sub-THz signals to users or act as boosters, forwarding signals along the stripe. Together with sub-10 GHz access points on the walls, they form the project’s “tandem” architecture, combining high data rates with robust coverage.
Users are served through beamforming, with Radio Units dynamically steering beams and handing over connections as users move. An intelligent algorithm continuously selects the best unit and beam, adapting to changing conditions.
The setup also highlights real-world challenges: sub-THz signals weaken over distance and can be blocked by obstacles. Here, the sub-10 GHz layer ensures reliable fallback coverage where high-frequency links struggle.
Compared to earlier methods, 6GTandem uses a smarter dual-frequency approach: combining sub-10 GHz signals with a few sub-THz pilots and machine learning to quickly predict the optimal connection.
Overall, the demonstrator shows how distributed sub-THz systems, intelligent coordination, and complementary lower-frequency support can enable scalable, high-performance indoor 6G networks—advancing Europe’s leadership in next-generation connectivity.

To reach this goal security research on real hardware is essential, as the architectural simulators—often the only option due to proprietary designs—don’t reflect the full complexity of actual hardware. This limits the ability to evaluate security changes or observe real hardware behavior.
To push security research on actual hardware, the ORSHIN team developed Proteus, an open-source RISC-V processor designed for rapid prototyping of security features. It includes tools for both security validation and performance testing.

Gas sensors are crucial in the personal and industrial monitoring to analyze personal exposure to air pollutants or to critical gases, to control product quality such as in the food industry, and in health care by analyzing gases from human body. These applications require miniaturized low power and low-cost gas sensors with good gas selectivity to be integrated in personal devices, in product packaging or in widely distributed sensor networks. AMUSENS aims at developing a gas sensor platform with flexible selectivity to different gas environments by combining a multi-pixel approach and artificial intelligence to adapt the data analysis to the targeted applications.

The MILADO project aims to revolutionize mid-infrared (MIR) laser technology by developing cost-effective, high-volume manufacturing processes for Quantum Cascade Lasers (QCLs). These lasers are essential for various spectroscopic applications due to their compactness, high power spectral density, and broad spectral coverage up to 12 µm. However, their high costs limit market penetration. MILADO merges "group III-V" and silicon photonics to create affordable, compact MIR light sources by using established complementary metal-oxide-semiconductor (CMOS) processes. This shift from expensive, complex craft production to scalable manufacturing could reduce QCL prices from thousands to tens of euros.

REALHOLO H2020 - Phase modulating micro mirror array for real holographic mixed-reality displays
The merging of real and virtual worlds to produce mixed reality (MR) environments is becoming a realistic component of future society. The effective use of MR demands a natural visual experience without physiological side effects for the user. REALHOLO is a project developed to meet these needs by researching advanced micro-mirror-based piston-type spatial light modulator (SLM) modulating the phase of visible light with optical features far superior to any liquid crystal-based alternative.The SLM will facilitate 3D display applications and active illumination and sensing.

Safe4RAIL-2 -CONNECTA-2: Final Dissemination Video
This video shows the impact and results from the Safe4RAIL-2 project which has examined ways of utilizing wireless (5G), interoperable, on-board communication as well as universal integration of the Train Control and Monitoring System (TCMS). In collaboration with its complementary action project CONNECTA-2, Safe4RAIL-2 has developed railway demonstrators where next-generation architectures and components for TCMS will be integrated. To eliminate costly field testing, Safe4RAIL-2 has performed testing in a simulated environment.
The results from Safe4RAIL-2 will define new train standards and pave the way for deterministic communications as well as secure and interoperable interconnections while increasing efficiency and safety of the TCMS.

We are halfway there - Phase 1 was completed successfully!
Our research and innovation action project Safe4RAIL-2 (Safe architecture for Robust distributed Application Integration in roLling stock 2) will look at ways of utilizing wireless (5G), interoperable, on-board communication as well as universal integration of the Train Control and Monitoring System (TCMS). In collaboration with its complementary action project CONNECTA-2 (826098), Safe4RAIL-2 aims to develop railway demonstrators where next-generation architectures and components for TCMS will be integrated. Another aspect of the project is to perform testing of the TCMS functions in a simulated environment. These test methods alleviate field-testing which would be costly to undertake.
The results from Safe4RAIL-2 will define new train standards and pave the way for deterministic communications as well as secure and interoperable interconnections while increasing efficiency and safety of the TCMS.

We bring hope to children with cancer
The project team will focus on identifying effective personalized medicine for paediatric cancers and will address a multitude of challenges. To meet these challenges, a comprehensive computational effort to combine knowledge base, machine-learning, and mechanistic models to predict optimal standard and experimental therapies for each child will be proposed. We will produce, assemble, standardize, and harmonize accessible high-quality multi-disciplinary data and leverage the potential of Big Data and HPC for the personalized treatments.
